In vitro hepatitis B virus infection of human bone marrow cells.

چکیده
Infection of humans with hepatitis B virus (HBV) frequently results in suppression of hematopoiesis; in some cases this may lead to severe bone marrow failure. The mechanism whereby HBV infection affects hematopoiesis is unknown. In vitro exposure of human bone marrow to HBV results in a dose-dependent inhibition of erythroid (erythroid burst forming units, BFU-E; erythroid colony-forming units CFU-E), myeloid (colony-forming units-granulocyte macrophage CFU-GM), and lymphoid (CFU-[T-lymphocytic]-TL) hematopoietic stem cells. Inactivation or immunoabsorption of HBV from sera resulted in loss of HBV-induced inhibition of hematopoietic stem cells. De novo gamma interferon was not detectable in the supernatants of cultures of bone marrow cells with HBV. Antibodies to gamma interferon did not affect the suppression of hematopoietic stem cells by HBV. Hepatitis B surface antigen (HBsAg) was detected by immune electron microscopy in nuclei of greater than 70% of immature hematopoietic cells including myeloblasts, normoblasts, and lymphoblasts; granulocytes had mostly cytoplasmic HBsAg. Hepatitis B virus core antigen (HBcAg) was also detected in about 5% of HBV infected bone marrow cells by immunoperoxidase staining. These data indicate that HBV can infect hematopoietic cells and their progenitors, thus suggesting a wider range of tropism for HBV than previously reported. These results may provide a basis to study HBV infection in vitro, and the effects of HBV on hematopoiesis.

Matrigel Enhances in vitro Bone Differentiation of Human Marrow-derived Mesenchymal Stem Cells
Objective(s) The use of co-culture cells as well as extra cellular matrix are among those strategies that have been employed to direct mesenchymal stem cell (MSC) bone differentiation in culture. In this regard, there is no study considering the effects of Matrigel on mesenchymal stem cell (MSC) in vitro bone differentiation. This was the subject of the present study. Materials and Methods ...

Abstract: Background and Aims: Huh-7 is a cell line that was derived from a liver tumor of a Japanese man. Hepatocellular carcinoma (HCC) is considered as a primary liver cancer. Highly resistant tumor to treatment which causes the death of many patients annually. Thus, targeting the cancer cells by using a new method could be effective in...
متن کاملEvaluation of In Vitro Differentiation of Cardiomyocyte-like cells Derived from Human Bone Marrow Mesenchymal Stem Cells
Purpose: To investigate the in vitro differentiation process of cardiomyocyte-like cells derived from human bone marrow mesenchymal stem cells under the influence of 5-azacytidine (5-aza). Materials and Methods: After purification, human bone marrow mesenchymal stem cells were exposed to 5-aza at a concentration of 5 μmol for 5 weeks to induce cardiomyocyte differentiation. To induce differenti...

Hepatitis B virus (HBV) is an etiological agent of hepatitis B infection. Hepatitis B is a life-threatening disease that affects the liver. The clinical outcomes of the disease are varied from asymptomatic disease to serious complication such as cirrhosis and hepatocellular carcinoma (HCC). Despite availability of the vaccine and appropriate treatment, hepatitis B infection still remains a majo...
متن کاملThe Molecular Prevalence of Viral Infections in Transplant Candidates with Bone Marrow Suppression, Shiraz, Southern Iran, 2010
BACKGROUND Transient bone marrow suppression, characterized by acute inability of the bone marrow to produce circulating blood cells, may strongly relate to the pathogenesis of some viral infections. OBJECTIVE To study the prevalence of some DNA and RNA viruses in patients with transient bone marrow suppression. METHODS EDTA-treated blood samples were collected from 27 patients with clinica...
